A comparison of colchicine treatment with a glycerine base and a water base.-
Two groups of seedlings of Vinca rosea (zn=16), 1000 in each group, were
subjected to treatment with a I percent colchicine-in-glycerine and I percent
colchicine-in-water, respectively. Two-thirds of each group were given five applications,
and one-third were given four applications by the brush method.
Approximately 5 percent of the plants treated with colchicine-in-glycerine
became tetraploid, whereas less than I percent tetraploidy resulted in those
treated with colchicine-in-water. A higher percentage of chimeras occurred in
the colchicine-in-glycerine treatment. There was a slightly greater mortality
of the plants subjected to the colchicine-in-water treatment. A I per cent solution
of colchicine-in-glycerine proved more effective for the induction of polyploidy
in Vinca. From a sampling of several hundred plants during the past
four years, no naturally occurring tetraploids have been detected.

Plant 100 plants.

Use the 1 percent colchicine-in-glycerine method.

Mix the two ingredients, and brush them onto your seedlings.

5 of these plants will be tetraploid.

If he read the thread, its covered earlier on. Three nodes occur randomly in diploids, triploids, and tetraploids.

A little review for those of you who didn't read the thread.

This is a trial run using very old pills (I have some fresh ones now).

The colchicine appears to have had some effects:
*Control group had a higher germ% and developed much quicker
*The treated groups showed some 'runts' and slow developers as well as 'normal' performers, although they were behind the control group

also

*The 'father' of all of the seed sets had 4 nodes in several sites (not a polyploid trait as it came from some good bagseed-none of the treated or control sprouts have displayed this trait as yet

Conclusions so far:
*I have seen nothing to indicate that any of the seedlings are polyploid
*Some or all of the seedlings may have aquired some polyploid traits that are not visible

I will be following these seedlings for a while to check development so as to be able to compare this set (old pills) with a future set using new pills.
I may even take them from veg to flowering if I have the room to do so.

I will be following the same procedure with the new pills when I do the 2nd expierement and will begin a new thread when I do so.

Read my post about doing 100 seedlings, and 5 of them will be polyploids. Its a 1% Glycerin-Colchicine mixture in water. I posted the study along with the post, and the person who did the study did 1000 plants and 50 of them turned out polyploids. He used the brushing method on them after the 3rd branch or so. I'd say do that and toss out all but the 1-5 polyploids you have and start cloning them.

I know most of you may not find this interesting but I want to record this for comparison when I do this again.

First 6 pics are of the 3 Sativa plants.
They look good, the first three sets of branches appear to be very close together. This holds true for most of the seedlings.

The next 2 pics are of seed set '3'
The last is set 'a'.
Being Indicas they appear to have broader leaves and are not quite as tall as the Sativas.

theman3000: I will be looking into the method you cite.
Not all that excited about applying the solution to the living plant but we will see. I have lots of time.

Colchicine is widely utilized in the botanical world, so you would think that there would be another way of getting it.
